PENGARUH LAMA PRAPERLAKUAN FLAVONOID RUTIN TERHADAP EFEK HIPOGLIKEMIK TOLBUTAMID PADA TIKUS JANTAN YANG DIINDUKSI ALOKSAN
Abstract
Rutin, a flavonoid is known as cytochrome P450 inhibitor that play role in
tolbutamid metabolism. In diabetic patients, oxidative stress is increased, so they
need antioxidant from outside of the body. Rutin use as dietary supplement which
has activities as antioxidant. The aim of this study is to know the influence of
Rutin againts hipoglycaemic effect of tolbutamid in alloxan-induced diabetic rats.
Fifteen male Sprague Dawley rats, aged about 2-3 months and weight
150-250 g which were induced by alloxan 150 mg/kgBW intraperitoneal (diabetic
rats), divided into five groups, each group contains 3 rats. Group I (control
diabetics) without treatment, group II was given CMC Na 1%, group III (positive
control) was given suspension of tolbutamid 360 mg/kgBW, groups IV and V were
given pretreatment of Rutin 45 and 180 mg/kgBW orally respectively an hour
before treatment of tolbutamid 360 mg/kgBW. Blood is taken from tail lateralis
vein at 60 minute before treatment, 30; 60; 120 and 180 minutes after treatment
of tolbutamid. The increased of data AUC30-180 from percentage blood glucosa
level was analysed by one way Anava and continued by LSD test with level of
confident 95%.
The results showed that pretreatment of Rutin dose 45 and 180 mg/kgBW
were not influenced the hypoglycaemic effect of tolbutamid 360 mg/kgBW
(p>0,05)
