Fragmentasi DNA Spermatozoa pada Tikus Jantan (Rattus Norvegicus) Diabetes Mellitus yang Diinduksi Streptozotocin Dapat Diturunkan dengan Pemberian Suspensi Bubuk Kedelai Kuning
Abstract
Diabetes mellitus disease is one of the degenerative diseases of the year rise and cause various complications. One complication is the distrupsion in the male reproductive system. Every effort began in earnest one with back to nature. Yellow soybean is one of alternative plants that can be utilized because of its content which is useful as anticarcinogenic, antioxidant, and antilipidemik antibiabetik. The aims of this study was to measure sperm DNA fragmentation levels of blood glucose, plasma MDA due to a suspension of powdered soy on male rat streptozotocin-induced diabetes mellitus. This research is purely experimental, post test only control group design on white male Wistar rats. The sample consisted of 30 male rats (Rattus norvegicus) aged 6-7 weeks weighing 150-300 grams were divided into five groups, namely K1: not induced with STZ and were not given a yellow suspension of soy powder, K2: STZ-induced and not given yellow soybean powder suspension, K3: given a dose of yellow soybean powder suspension 200 mg/kg/day after STZ-induced hyperglycemia and occurs, K4 is given a dose of yellow soybean powder suspension 400 mg/kg/ day after STZ-induced hyperglycemia and occur, given K5 yellow soybean powder suspension dosage 800 mg/kg/day after STZ-induced and occurs hiperglikemi. Before and after treatment was examined for glucose and MDA, the end of the treatment of sperm retrieved from cauda epididymis and was examined for DNA fragmentation by using acridine orange staining. The results of data analysis, ANOVA and t-test showed that there were differences in mean blood glucose, MDA and a good percentage of spermatozoa between groups (p = 0,000). The mean sperm DNA fragmentation in STZinduced diabetic rats fed soybean powder yellow suspension was lower than that which is not given a yellow soybean powder suspension. Giving a yellow suspension of soy powder can lower blood glucose levels and MDA significantly in streptozotocin-induced diabetic rats
