dc.description.abstractHasil berbagai penelitian menunjukkan bahwa kanker masih merupakan penyebab utama kematian di Indonesia maupun di dunia. Penggunaan kombinasi kemoterapi, penggunaan bersama senyawa kemoprevensi yang bersifat non-toksik atau lebih tidak toksik dengan agen kemoterapi, diketahui mampu meningkatkan sensitifisitas sel kanker serta efikasi kemoterapi dengan penurunan toksisitas terhadap jaringan normal. Penelitian ini bertujuan mengetahui potensi kombinasi ekstrak etil asetat daun dewandaru (Eugenia uniflora L.) dan Doxorubicin dalam menghambat proliferasi sel kanker payudara T47D secara in vitro dengan penentuan nilai combination index-nya. Uji MTT digunakan untuk mengukur besarnya efek penghambatan sel T47D oleh doxorubicin dan ekstrak etil asetat daun dewandaru. Nilai IC50 ekstrak etil asetat E. uniflora adalah 212,79 µg/ml yang berarti ekstrak tersebut memiliki efek sitotoksik yang kurang poten terhadap sel T47D, karena nilai IC50 lebih besar dari 100 µg/ml. Hal ini sesuai dengan hasil penelitian sebelumnya. Nilai IC50 doxorubicin secara tunggal pada sel T47D yang diperoleh pada penelitian ini adalah 103,12 nM. Hasil ini berbeda dengan hasil penelitian sebelumnya yang menunjukkan nilai IC50 doxorubicin sebesar 50 nM, di bawah 100 nM. Efek kombinasi dox dan ekstrak etil asetat daun dewandaru pada sel T47D belum dapat ditetapkan karena kegagalan penentuan prosentase sel hidup dengan metode MTT.en_US
