EFEK PENGHAMBATAN BUNGA ROSELLA MERAH ( Hibiscus sabdariffa Linn) SEBAGAI ANTI ATEROSKLEROSIS TERHADAP AKTIFASI NF-κβ PADA KULTUR SEL ENDOTHEL YANG DIPAPAR LOW DENSITY LIPOPROTEIN TEROKSIDASI
Abstract
Background The atherosclerosis disease has recently been a global threat.
Seeing the aterosclerosis pathomechanism, Ox-LDL was one of major source of
aterogenesis process through the ROS formation as among other which would activate
NF-κβ and leading to the occurrence of inflammation in aterosclerosis disease. Rosella
(Hibiscus sabdariffa Linn) was considered having effect on this track, but it still need to
be proven together with its dosage. Recent studies indicate that Rosella is a tropical
plant containing antioxidant, that is polyphenol compound (protocatechuic acid and
antocyanin) and ascorbate acid invitro and invivo, and is widely distributed in Indonesia.
The work mechanism of the red flower on the aterogenesis inhibition has not been
largely known.This study aimed to investigate whether Red Rosella Flower extract
inhibit NF- κβ activation that mediated the occurrence of inflammation on
atherosclerosiby using Ox-LDL-treated HUVECs.
Materials and methods Cultured endothelial cell (HUVECs), is used for a
model of endothelial cells. On 5 treated groups, that was the group of HUVECs culture
without presenting Ox-LDL as negative control; groups with 40 ug/ml Ox-LDL
presentation (dosage was according to exploratory research) as positive control; groups
given the Rosela flower abstract with various dosages (0.001 mg/ml, 0.005 mg/ml and
0.01 mg/ml) then being presented Ox-LDL. 40 ug/ml of Ox-LDL is treated on HUVECs
to stimulate responds of endothelial cells mimicking dyslipidemia condition. The Red
Rosella flowerin a various dose, and a vehicle for control, were given 2 h before Ox-
LDL treatment. The NF-κβ activaton measured by immunohistochemistry.
Result The red rosella flower treatment on Ox-LDL-treated HUVECs, in
adequate dose, may result in prevention NF-κβ activation of in respond to Ox-LDL
treatment, is (0.001 mg/ml, 0.005 mg/ml and 0.01 mg/ml) dosages. By correlation
Spearman`s (p=0.01, Correlation Coeff= -1), there is negative correlation between NF-
κβ activation in positive control groups and groups given the Red Rosela tea extract
with various dosages.This study may contribute a benefit for prevention and medication
for atherosclerosis disease progression in the future, particulary inhibition of NF-κβ
activation. The red Rosella flower with 0.001 mg/ml or 0.048 g dried calycx can use
consumption of diet.
