DAYA HAMBAT EKSTRAK DAUN DEWANDARU (Eugenia uniflora L.) TERHADAP AKTIVITAS GLUTATION S-TRANSFERASE PARU TIKUS SECARA IN VITRO
Abstract
Objective: In some tumor cases, the Glutathione S-Transferase (GST)
have been known increased. It causes anticancer drugs would be metabolized
faster. Dewandaru leaves extracts were shown inhibitory effect on GST rat liver
activity.This research aims are to determine the inhibitory potency of chloroform,
ethyl acetate, and ethanol extract of Dewandaru leaves on GST activity of rat
pulmo by in vitro using 1-chloro-2,4-dinitrobenzene (CDNB) as substrate.
Methods: The research was performed by using cytosolic fraction of rat pulmo
which is prepared by centrifugation. Chloroform, ethyl acetate, and ethanol
extracts of Dewandaru leaves were obtained by maceration. The protein content of
cytosolic fraction was measured by Lowry method using bovine serum albumin
(BSA) as standard. The GST activity was known from the conjugated product
between Glutathione (GSH) and the substrate. It was measured by
spectrophotometer. IC50 of each extract was determined from the plots of
remaining activities versus the (varied) concentration of extract at a fixed
concentration of substrate.
Results: The research results showed all extracts have inhibitory effect on GST
activity. IC50 values of chloroform, ethyl acetate, and ethanol extract using CDNB
as a substrate are 77.964; 299.305; 160.014 µg/ml respectively.
Conclusions: Dewandaru leaves extracts were shown inhibitory effect on GST rat
pulmo activity by in vitro using CDNB as substrate.
