Kluthuk banana (musa balbisiana colla), a seeded banana, has been studied for its ability to reduce gastric acid
secretion. Is the active compound hydrophylic or hydrophobic has not been known yet.The aim of his study is to
determine the effect of the seed of ripe kluthuk banana (musa balbisiana colla) etheric and ethanolic extract, and to
compare the effect of both extract on rat gastric acid secretion in vitro. This study was conducted using isolated
Wistar rat stomach, according to the methods modified from Barocelli. Fifty four rats (3-4 month) of 150-250 g were
used in this study. They were divided into 9 groups (6 rats each). The rats were fasted and drinking water was given ad
libitum for 24 hours before testing. The rats were weighted, anesthetized with ether inhalation, and then sacrified.
The stomach was taken and suspended in an organ bath containing 37°C buffered serosal solution and bubbled
with carbogen. The gastric lumen was perfused continuously with unbuffered mucosal solution 1 ml min-1 and
bubbled with 100% O2. The isolated preparation was stabilized for 1 hour and perfusate spilled out. The perfusate
+ was allowed to flow continuously, and collected for 10 minutes duration. The H consentration was measured by
+ titration with NaOH 0,002 N using phenolftalein as indicator, known as basal H consentration . Pretreatment was
added to the unbuffered mucosal solution for 30 minutes, after which the treatment (histamine 10-6 M) was added
to the unbuffered solution in each group. Perfusate from gastric lumen were collected every 10 minutes untill 80
+ + minutes and H consentration were measured by mean of titration. The H consentration elevation was expressed
as mean ± SEM, the AUC0-80 was calculated and analyzed by ANOVA.The result showed that the total area under
+ curve (AUC0-80) of H consentration was 14550 ± 692,70 in control group (saline), 141,67 ± 2838,62 in etheric
extract of the seed of ripe kluthuk banana doses-1 group (EESRKB I group) equivalent to 1,92 mg/kgBW, 8516,67 ±
3659,64 in EESRKB II group (equivalent to 3,84 mg/kgBW), 5650 ± 3191,94 in EESRKB III group (equivalent to 7,69
mg/kgBW), 4500 ± 2819,22 in ethanolic extract of the seed of ripe kluthuk banana doses-1 group (AESRKB I group)
equivalent to 8,7 mg/kgBW, -5883,33 ± 760,45 in AESRKB II group (equivalent to 17,4 mg/kgBW), -1116,67 ± 3444,36
in AESRKB III group (equivalent to 34,8 mg/kgBW), -1333,33 ± 80,28 in cimetidin group, and 12683,33 ± 968,65 in
DMSO group. The statistical analysis showed that the AUC0-80 of EESRKB and AESRKB were significantly (p<0,05)
lower than those of control solution (except EESRKB II), the AUC0-80 of EESRKB I were not significantly different
(p>0,05) than those of AESRKB I, the AUC0-80 of EESRKB II were significantly higher (p<0,05) than those of AESRKB II,
and the AUC0-80 of EESRKB III were not significantly different (p>0,05) than those of AESRKB III. It was concluded
that the etheric and ethanolic extract of the seed of ripe kluthuk banana (Musa balbisiana Colla) showed an
inhibitory effect on rat gastric acid secretion induced by histamine, and the ethanolic extract showed an inhibitory
effect more than those of etheric extract.
