dc.description.abstractPenelitian awal menunjukkan efek sitotoksik ekstrak etanol rimpang lempuyang gajah (Zingiber zerumbet) dan lempuyang emprit (Zingiber littorale) terhadap sel kanker payudara (T47D) dan identifikasi zerumbone sebagai senyawa mayor secara KLT. Pengembangan obat herbal sebagai antikanker payudara memerlukan penelusuran mekanisme molekuler sehingga menghasilkan obat yang selektif pada sel kanker dan aman. Selain itu, kontrol kualitas obat herbal perlu dilakukan melalui standarisasi ekstrak, penentuan kadar senyawa penanda dan profil metabolitnya. Penelitian pada tahun pertama meliputi: (1). Pembuatan ekstrak (2). Standarisasi ekstrak (parameter spesifik dan non spesifik) (3). Kontrol kualitas ekstrak berbasis senyawa penanda zerumbone dengan validasi metode KCKT dan analisis profil metabolit dengan KG dan (4). aktivitas antikanker payudara terhadap sel T47D dengan pengamatan morfologi sel dan apoptosis. Tahun kedua meliputi: (1). Uji toksisitas akut dan (2). Uji toksisitas subkronis. Tujuan penelitian tahun pertama adalah mengembangkan metode kontrol kualitas ekstrak Z. zerumbet dan Z. littorale berbasis senyawa penanda zerumbone dan aktivitas antikankernya. Tahun kedua ditujukan untuk pengembangan formulasi dan desain produk OHT serta pengujian toksisitas akut-subkronisnya. Target luaran khusus dari penelitian ini adalah metode kontrol kualitas ekstrak terstandar (tahun pertama) dan ekstrak Z. zerumbet dan Z. littorale sebagai obat antikanker yang selektif, aman dan bermutu (tahun kedua). Ekstrak lempuyang emprit dan lempuyang gajah sudah memenuhi persyaratan parameter non spesifik dan spesifik. Kadar zerumbone pada ekstrak lempuyang emprit berbeda untuk setiap daerah (18,24 s/d 27,24 %b/b), dan kadarnya pada ekstrak lempuyang gajah jauh lebih bervariasi untuk setiap daerah (8,37 s/d 32,31 %b/b). Profil metabolit dari ekstrak lempuyang emprit dan lempuyang gajah berbeda pada setiap daerah, dengan kandungan metabolit mayornya berupa asam-asam organik dan zerumbone. Ekstrak lempuyang gajah memiliki aktivitas sitotoksik terhadap sel T47D yang jauh lebih kuat (IC 50 emprit (IC 50 > 50 μg/mL). < 50 μg/mL) dibandingkan ekstrak lempuyangin_ID
